چکیده

A significant number of bacteria, including the emerging multidrug-resistant “superbugs,” such as Pseudomonas aeruginosa, Clostridium difficile, Staphylococcus aureus, and Escherichia coli, secrete highly potent exotoxins with no antitoxins currently available to disable them. Several bacterial toxins, such as the botulinum neurotoxin (BoNT) of Clostridium botulinum, can be aerosolized and used as biological weapons directly. With some bacterial diseases, such as anthrax, flu-like symptoms appear only after bacteria have proliferated inside the host and have started to produce toxins that ultimately cause death [1]. Even though aggressive antibacterial treatment effectively inhibits Bacillus anthracis propagation, patients can still die because of intoxication [2]. Furthermore, several engineered strains of B. anthracis, which are resistant to the existing antibacterial agents, have already been developed [3,4]. Therefore, the discovery and development of small-molecule antitoxins represent a high-priority task in drug design, medicinal chemistry, and biodefense [4,5]. In the last ten years, a number of biophysical studies have focused on finding ways to disable those bacterial toxins which form ion channels in the host’s cell membranes. This chapter focuses on the existing efforts to design antitoxins that counteract channelforming bacterial toxins.

DBETH: A Database of Bacterial Exotoxins for Human

Pathogenic bacteria produce protein toxins to survive in the hostile environments defined by the host's defense systems and immune response. Recent progresses in high-throughput genome sequencing and structure determination techniques have contributed to a better understanding of mechanisms of action of the bacterial toxins at the cellular and molecular levels leading to pathogenicity. It is fa...

Expression of a Chimeric Protein Containing the Catalytic Domain of Shiga-Like Toxin and Human Granulocyte Macrophage Colony-Stimulating Factor (hGM-CSF) in Escherichia coli and Its Recognition by Reciprocal Antibodies

Fusion of two genes at DNA level produces a single protein, known as a chimeric protein. Immunotoxins are chimeric proteins composed of specific cell targeting and cell killing moieties. Bacterial or plant toxins are commonly used as the killing moieties of the chimeric immunotoxins. In this investigation, the catalytic domain of Shiga-like toxin (A1) was fused to human granulocyte macrophage ...

In silico analysis of protein toxin and bacteriocins from Lactobacillus paracasei SD1 genome and available online databases

Lactobacillus paracasei SD1 is a potential probiotic strain due to its ability to survive several conditions in human dental cavities. To ascertain its safety for human use, we therefore performed a comprehensive bioinformatics analysis and characterization of the bacterial protein toxins produced by this strain. We report the complete genome of Lactobacillus paracasei SD1 and its comparison to...

Channel-Forming Bacterial Toxins in Biosensing and Macromolecule Delivery

To intoxicate cells, pore-forming bacterial toxins are evolved to allow for the transmembrane traffic of different substrates, ranging from small inorganic ions to cell-specific polypeptides. Recent developments in single-channel electrical recordings, X-ray crystallography, protein engineering, and computational methods have generated a large body of knowledge about the basic principles of cha...
